How do you calculate protein fold-change (Western)?

Loading-control-normalized densitometry: relative density and fold change vs control. The relationship used is fold = (target/loading) / mean(control target/loading).

  • Control loading: 100, 100

Formula: fold = (target/loading) / mean(control target/loading)

The working

  1. Relative density: target / loading control = per sample
  2. Control mean: mean of "control" = 1.1
  3. Fold change: rel. density / control mean = per sample

Treated = 2× ± 0.182 (SEM, n=2)
